F0 crispant screening approach identifies nrf2a as an actor of late thyroid development in zebrafish.
Live imaging of transgenic Tg(tg:nlsEGFP) zebrafish embryos from 55 to 144 hpf after one-cell-stage injection of single-guide RNA and Cas9 protein targeting either nrf2a, nrf2b, or admatsl2. Epifluorescence live imaging showed that early stages of thyroid development are not affected in nrf2a and nrf2b crispant embryos compared with their adamtsl2 crispants and WT siblings. However, at 6 dpf nrf2a crispants displayed enlargement of their thyroid glands compared with their nrf2b and adamtsl2 crispants and noninjected siblings.
