Fig 5
polr3asa907/sa907 zebrafish exhibit globally reduced proliferation and increased cell death.
TUNEL staining for cell death (magenta) and pH3 staining for proliferation (white) from 3–5 dpf were quantified throughout the cranial region and normalized to DAPI (blue, outlined). (A, B) At 3 dpf, increased cell death was observed in the tectum and eye along with a slight decrease in proliferation. TUNEL is significantly increased (C; *p = 0.03) while pH3 is significantly decreased (D; *p = 0.01). n = 12 controls, n = 12 mutants. (E,F) At 4 dpf, cell death remains increased (G; **p = 0.0098), and pH3 was slightly reduced, but this was not statistically significant (H; ns, p = 0.27). n = 12 controls, n = 12 mutants. (I,J) By 5 dpf TUNEL remains significantly increased (K; **p = 0.005; n = 18 controls, n = 17 mutants) and pH3 is significantly reduced (L; *p = 0.01; n = 10 controls, n = 10 mutants). Scale bar = 100 µm.
