Fig 4 Cell death is increased within the craniofacial cartilage in polr3asa907/sa907 zebrafish.
Cell death (TUNEL, magenta) and proliferation (pH3, white) were quantified within pharyngeal arch 1 and 2 derived cartilage elements using sox10:egfp (green, outlined) to mark this domain. (A,B) At 3 dpf, there is a significant increase in cell death (C; *p = 0.016), but no change in proliferation (D; ns, p = 0.51). n = 12 controls‌‌, 12 mutants. (E,F) At 4 dpf, differences in the morphology of the craniofacial cartilage are visible, including a reduced basihyal, while cell death (G; ns, p = 0.08) and pH3 remain unchanged (H; ns, p = 0.94). n = 12 controls, 12 mutants. (I,J) By 5 dpf, there is a noted difference in cartilage morphology and increased cell death (K; **p = 0.008; n = 17 controls, 17 mutants), while pH3 is unchanged (L; ns, p = 0.48; n = 9 controls, 10 mutants), suggesting that differences are due to changes in cell death.
